Quantitative Analysis of 3D Microtissue Growth and Biomarker Intensity

ثبت نشده
چکیده

In vivo cells form three-dimensional (3D) microenvironments through cell-cell and cell-matrix interactions to create complex tissues and perform higher level functions. Biochemical signals and nutrient gradients are essential for tissue functioning and therefore also physiological cell behavior. Conventional two-dimensional cell culture does not provide such a complex microenvironment and consequently cells can lose some of their physiological properties (Pamploni et al., 2007). 3D cell culture methods are now widely accepted as being more physiologically relevant and are believed to improve prediction of drug candidates at an earlier stage of the development process (Fayad et al., 2011, Hirschaeuser et al., 2010).

High-content analysis of biomarker intensity and distribution in 3D microtissues

Three-dimensional (3D) cell culture methods are widely accepted as more physiologically relevant than conventional 2D cell culture methods and are believed to improve the prediction of drug candidates at an early stage of the drug development process. Here we describe the analysis of a spherical colon cancer microtissue model using the Operetta® High Content Imaging System. In vivo near-infrare...

Multiplexed, high-throughput analysis of 3D microtissue suspensions.

Three-dimensional (3D) tissue models have significantly improved our understanding of structure/function relationships and promise to lead to new advances in regenerative medicine. However, despite the expanding diversity of 3D tissue fabrication methods, approaches for functional assessment have been relatively limited. Here, we describe the fabrication of microtissue (μ-tissue) suspensions an...

3D-Conformal Radiation Therapy and Intensity-Modulated Radiation Therapy Techniques for Laryngeal Cancer Taking Parotid Glands as Organ at Risk

Background: Three-dimensional conformal radiotherapy (3D-CRT) and intensity-modulated radiotherapy (IMRT) techniques are used for the treatment of patients with laryngeal cancer. Objective: This study aimed to investigate the effects of these 2 treatment techniques on the planning target volume (PTV) (laryngeal cancer), dose homogeneity, dose of organs at...

Three-Dimensional Finite Element Analysis of Stress Intensity Factors in a Spherical Pressure Vessel with Functionally Graded Coating

This research pertains to the three-dimensional (3D) finite element analysis (FEA) of the stress intensity factors (SIFs) along the crack front in a spherical pressure vessel coated with functionally graded material (FGM). The vessel is subjected to internal pressure and thermal gradient. The exponential function is adopted for property of FGMs. SIFs are obtained for a wide variety of crack sha...
